千坂先生研究内容紹介
- 研究名 Cornichon-like protein facilitates secretion of HB-EGF and regulates proper development of cranial nerves.

末梢への移動過程において、頚部神経堤細胞は菱脳分節3、5の側方間充織でErbB4依存的に排斥される。ErbB4のリガンドは多く、実際にどのリガンドが上記機構に関与するか不明であった。今回我々はコルニション類似タンパクがニワトリ胚菱脳分節3、5に発現することを発見し、培養細胞系ではコルニション類似タンパクはErbB4リガンドの一つであるHB-EGFの分泌を促進することを明らかにした。カルボキシ末端を欠いたコルニション類似タンパクをニワトリ胚で強制発現すると神経堤細胞の分布が異常になり、鰓弓神経節間に異常な軸索連絡が生じる。siRNAによるコルニション類似タンパクやHB-EGFの発現抑制でも類似の表現型が見られた。HB-EGFは菱脳では広範囲に発現しており、コルニション類似タンパクはHB-EGFの機能をErbB4とともに菱脳分節3、5に限局する働きをしていると思われる。
